Computer Interfacing
Enzyme Action: Testing Catalase Activity
Prepare the computer for data collection.
Load an Experiment File:
If you have Biology with Computers, open "EXP04.LXP"
from the experiment files of Data Logger. That's it! The computer
should be ready to collect data.
Make an Experiment File:
If you do not have Biology with Computers, set the
Data Logger program as follows:
From the Display menu
- Choose One Graph
- Choose Labels & Units.
- Enter Transmittance for the label of Port 1.
- Enter T1 for the short name
- Enter % for the units.
From the Data menu
- Choose New Column
- In the Formula text box, type 2-log("T1")
- In the Full Name text box, type Absorbance.
- In the Short Name text box, type A.
- Click on the OK button.
- Choose New Column again.
- In the menu near the right top (which will display "Formula"),
select Prompted.
- In the Full Name text box, type Concentration.
- In the Short Name text box, type Conc.
- In the Units text box, type %
- Click on the OK button.
Alter the plot axes
- Select Absorbance for the vertical axis. To do this, click
on the vertical axis title and hold the mouse button down. A menu
will pop up. Select Absorbance from this menu.
- The vertical axis has Absorbance scaled from 0 to 1. To change
the plot labels:
- Double click the mouse when the cursor is over the lowest
value displayed on the vertical axis of the plot. When
the value is highlighted, type "0".
- Double click the mouse over the highest value
displayed on the vertical axis of the plot. When the value is
highlighted, type "1".
- The horizontal axis has Conc scaled from 0 to 100%.
To change the plot labels:
- Double click the mouse when the cursor is over the lowest
value displayed on the horizontal axis of the plot. When
the value is highlighted, type "0".
- Double click the mouse over the highest value
displayed on the horizontal axis of the plot. When the value is
highlighted, type "100".
From the Collect menu
- Turn the Event Mode on, if necessary. Examine the Event Mode
menu item. If it is not checked, choose it to select the menu
item.
- Turn the Display Inputs on, if necessary. Examine the Display
Inputs menu item. If it is not checked, choose Display Inputs
to select it.
- If only one colorimeter is attached to the interface, choose
Select Inputs. Un-check the box next to "Probe in Port 2".
It is possible for two groups to use one interface, provided they
work well together! The two sets of experiments must by synchronized
for this to happen. If two colorimeter are attached to the interface,
choose Select Inputs. Check the box next to "Probe in Port
2".
